法律分析:
辞职申请书和辞职信两者的根本区别，就是用人单位收到辞职信后给劳动者办理辞职手续时不必支付经济补偿金。而用人单位在收到辞职申请书后做出同意的意见或者盖章，则视为双方协商达成一致解除劳动合同，用人单位必须支付经济补偿金。
法律依据:
《中华人民共和国劳动合同法》
第三十六条 用人单位与劳动者协商一致，可以解除劳动合同。
第三十七条 劳动者提前三十日以书面形式通知用人单位，可以解除劳动合同。劳动者在试用期内提前三日通知用人单位，可以解除劳动合同。
